PSMG3 antisense RNA 1 (PSMG3-AS1) is a long non-coding RNA that does not encode protein but regulates gene expression at transcriptional and post-transcriptional levels[2][4]. PSMG3-AS1 is upregulated in multiple cancer types, including oral squamous cell carcinoma, lung squamous cell carcinoma, breast cancer, and glioblastoma[1][3][4][6]. It acts primarily by interacting with specific microRNAs such as miR-141 and miR-143-3p, functioning as a competitive endogenous RNA ("miRNA sponge"). Through such mechanisms, it promotes oncogenic processes, notably increasing cell proliferation, migration, and invasion, and is associated with poor patient prognosis in several cancers[1][3][4][6]. In LUSC, high expression correlates with metastatic potential and worse overall survival, making PSMG3-AS1 both a potential therapeutic target and a biomarker for prognosis[4]. Its capacity to regulate oncogenic signaling pathways and affect the maturation/function of miRNAs highlights its potential as a novel long non-coding RNA cancer biomarker and therapeutic target, though no known drugs directly target it at present[6].
