Overview

PTEN-induced kinase 1 (PINK1) is a mitochondrial serine/threonine-protein kinase that plays a critical role in protecting cells from stress-induced mitochondrial dysfunction and is directly involved in mitochondrial quality control, particularly through mitophagy. It phosphorylates key substrates such as ubiquitin and Parkin’s ubiquitin-like domain to activate Parkin E3 ligase activity, which triggers tagging of damaged mitochondria for autophagic degradation. Mutations in PINK1 cause autosomal recessive early-onset Parkinson's disease.
