Overview

Pteridine reductase 1 (PTR1) is an NADPH-dependent enzyme found in Leishmania species, protozoan parasites responsible for leishmaniasis. PTR1 plays a crucial role in the salvage and reduction of pteridines, compounds essential for parasite growth and survival, because Leishmania are auxotrophic for these molecules. Inhibition of PTR1 is a potential therapeutic strategy, but the redundancy with DHFR means dual inhibition may be required. Upregulation of PTR1 confers resistance against antifolate drugs.
