PTPRG antisense RNA 1 (PTPRG-AS1) is a long non-coding RNA (lncRNA) transcribed on the opposite strand of the protein tyrosine phosphatase receptor type G (PTPRG) gene, and is not a protein-coding target but a regulatory RNA molecule[2]. It is upregulated in various types of cancer, including non-small cell lung cancer (NSCLC), lung adenocarcinoma, and epithelial ovarian cancer, where it acts as an oncogene[1][3][4][6]. PTPRG-AS1 promotes cancer cell proliferation, cell cycle progression, migration, invasion, and radioresistance. Mechanistically, PTPRG-AS1 functions as a competing endogenous RNA (ceRNA), sponging tumor-suppressive microRNAs such as miR-200c-3p, miR-124-3p, and miR-545-3p, thereby derepressing oncogenic proteins such as TCF4, cyclin D1 (CCND1), and HDAC4[1][3][4][5]. Silencing or downregulating PTPRG-AS1 inhibits cancer cell growth, induces apoptosis, and may enhance radiosensitivity, suggesting a potential prognostic or biomarker role, though it is not currently a direct therapeutic target nor are drugs known to specifically modulate it[1][2][4][5].
