Overview

Pumilio RNA binding family member 3 (PUM3) is a divergent member of the PUF (Pumilio and Fem-3 binding factor) protein family, which are conserved RNA binding proteins known primarily for post-transcriptional regulation of mRNA stability and translation[2][6][7]. Unlike classical PUF proteins, PUM3 is predominantly localized in the nucleolus where it participates in ribosomal RNA processing, specifically in the conversion of 7S to 5.8S rRNA, and is critical for 5.8S small ribosomal subunit assembly[2][6]. PUM3 contains an extended RNA-binding domain with 11 Pumilio repeats, giving it distinct RNA structural recognition properties and pre-rRNA processing specificity[2][6]. PUM3 can also bind DNA and has been shown to inhibit poly(ADP-ribosyl)ation activity and degradation of PARP1 after genotoxic stress[4]. Altered expression of PUM3 has been associated with cancer, autoimmunity, and learning disorders, but it is not currently considered a direct therapeutic target and there are no known drugs specifically acting through this molecule[2][6][7].
