Purine-rich box 1 (PU.1) is a critical ETS-family transcription factor essential for hematopoiesis and immune cell differentiation. It binds to purine-rich sequences (PU-box) on enhancers of target genes, regulating their expression by recruiting cofactors and chromatin remodelers. PU.1 plays indispensable roles in myeloid lineage development, B-cell lineage specification, and dendritic cell development. Dysregulation or mutation can contribute to leukemogenesis and other diseases.
PU.1 binds to purine-rich motifs on enhancers, regulating gene expression by interacting with cofactors and chromatin remodelers. It acts as an architectural transcription factor, inducing DNA bending to promote the binding of other transcription factors.
