Overview

Putative homeodomain transcription factor 2 (PHTF2) is an evolutionarily conserved protein, classified as a transcription factor, and is encoded by the PHTF2 gene in humans[5][7]. Although referred to as a “putative” transcription factor due to sequence homology to homeodomain proteins, its detailed functions remain incompletely characterized. PHTF2 shows DNA-binding transcription factor activity[7], and it has been identified as an oncogenic factor in several studies. Notably, it is upregulated in various cancers, including lung adenocarcinoma and gastric cancer, where it correlates with tumor progression, metastasis, and poor patient prognosis[1][2][4]. Functional studies implicate PHTF2 in regulating cell proliferation, migration, invasion, and intracellular lipid metabolism, as well as possibly modulating mitochondrial efficiency and stress responses[1][2][3]. It has emerged as a hub gene in protein-protein interaction analyses of differentially expressed genes in cancer, underscoring its putative importance in oncogenic networks[2]. Despite growing interest, there are currently no targeted drugs or established clinical interventions specifically modulating PHTF2 function.
