PIPSL is a transcribed pseudogene showing similarity to two adjacent loci on chromosome 1: phosphatidylinositol-4-phosphate 5-kinase type I alpha (PIP5K1A) and proteasome 26S subunit, non-ATPase, 4 (PSMD4). It likely arose by L1 retrotransposition of a read-through transcript combining these two loci into chromosome 10. Although transcriptional activity has been reported, there is no strong evidence that a functional protein is produced or translated. The predicted protein has negligible phosphatidylinositol phosphate kinase activity and can bind ubiquitinated proteins, but these functions are not considered biologically significant. Genomic databases (RefSeq, UniProt, GeneCards) consistently classify PIPSL as a pseudogene and not a validated therapeutic target. The locus is tightly regulated and testis-specific in primates but is not considered an active receptor, enzyme, transporter, or drug target. PIPSL should not be considered a valid drug target. Its classification as a pseudogene means it does not produce functional protein relevant to current therapeutic applications. Any use of PIPSL as a therapeutic target would be incorrect.
