The Puumala virus (PUUV) glycoproteins are essential structural and functional components of the hantavirus envelope. The two main glycoproteins, Gn and Gc, are derived from a precursor polyprotein encoded by the M segment of the viral genome. These proteins play critical roles in virus attachment, entry into host cells, membrane fusion, and immune recognition. The Gn/Gc complex forms hetero-tetrameric spikes on the virion surface that mediate initial attachment to host cell receptors. Gc is a class II membrane fusion protein that undergoes conformational changes at low pH within endosomes to drive fusion between viral and cellular membranes. Both proteins are primary targets for neutralizing antibodies; their structure determines antigenicity and immune evasion potential.
