PVRIG (PVR Related Immunoglobulin Domain Containing), also known as CD112R (CD112 receptor), is an inhibitory immune checkpoint receptor primarily expressed on T cells and NK cells. It interacts with Nectin-2/CD112 (PVRL2), leading to suppression of T-cell function and regulation of NK cell activity. PVRIG is a target for cancer immunotherapy, and blocking its interaction with PVRL2 can enhance anti-tumor immune responses, potentially synergizing with other immunotherapies such as PD-L1 blockade.
