Overview

PWWP domain-containing DNA repair factor 3A (PWWP3A) is a chromatin-associated protein characterized by the presence of a PWWP domain, which enables recognition of methylated histone H3 lysine 36 (H3K36me2/3) marks[6]. PWWP3A facilitates the maintenance and remodeling of chromatin architecture, especially in response to DNA damage[1][5][7]. It is rapidly recruited to sites of DNA breaks through interaction with the DNA repair protein TP53BP1, promoting localized chromatin relaxation and efficient repair, and is important for cell survival after genotoxic stress[3][4][5][7]. In animal studies, PWWP3A is highly expressed in the testis and is implicated in the aging-associated degeneration of this tissue, potentially due to defects in DNA repair or immune regulation[6]. There is no current evidence of targeting PWWP3A with clinical drugs, nor is it widely established as a biomarker or direct therapeutic target[1][3][4][7].
