Overview

PWWP domain-containing DNA repair factor 3B (PWWP3B) is a protein characterized by the presence of a PWWP domain, a conserved motif found in several chromatin-associated proteins. The PWWP domain mediates recognition of specific histone methylation marks—most notably trimethylation of lysine 36 on histone H3 (H3K36me3)—and allows the protein to interact with both DNA and nucleosomes, often to regulate transcription and chromatin state[1][3][5]. PWWP3B is also known as MUM1L1 (Mutated melanoma-associated antigen 1-like 1) and is part of a family whose members have been implicated in DNA repair and chromatin regulation[2][4]. While closely related PWWP3A has defined roles in DNA damage response and innate immune regulation, current evidence suggests that PWWP3B does not share redundancy in antiviral signaling pathways[2]. The gene encoding PWWP3B is expressed in certain tumors, and its domain structure suggests potential roles in cancer biology and epigenetics, but specific molecular and therapeutic targeting information is limited[4].
