PWWP domain-containing protein 2A (PWWP2A) is a chromatin-binding adapter protein involved in multiple epigenetic and transcriptional control mechanisms. It binds nucleosomes by recognizing specific histone marks, primarily histone H3K36me3 and the variant H2A.Z, and recruits chromatin-modifying complexes that regulate histone acetylation at active genes[1][3]. This process plays a crucial role in normal gene expression, chromatin structure, DNA repair, and mitotic progression. PWWP2A is essential for proper control of transcriptional elongation and suppression of aberrant transcription initiation. Mutations or altered expression of PWWP2A are linked to several diseases, notably Ivic syndrome and certain cancers such as Spitzoid melanoma[1][2][3]. It is found primarily in the cell nucleus and is part of epigenetic regulatory complexes, including interactions with the NuRD subcomplex[1]. Notes: - PWWP2A is not currently recognized as a classic "therapeutic target" (e.g., receptor, enzyme, transporter), but as an epigenetic regulatory protein with emerging connections to disease. There are no known drugs directly targeting PWWP2A as of the current literature[1][2]. - Due to its role in cancer and gene regulation, future targeting in drug discovery is possible, but as of now, relevant drugs, mechanisms of drug action, and biomarker or safety information are not established.
