Overview

PWWP domain-containing protein 2B (PWWP2B) is a nuclear chromatin-binding protein characterized by its PWWP domain, an approximately 70 amino acid motif mediating protein-protein and protein-DNA/histone interactions. It forms a variant NuRD complex that modulates histone acetylation at actively transcribed genes and affects transcription elongation via RNA polymerase II. PWWP2B also plays a crucial role in DNA double-strand break repair, being rapidly recruited to damage sites through interactions with UHRF1 and MRE11, facilitating homologous recombination and genome integrity. Mutations in the gene are recurrent in gastric adenocarcinoma and associated with poor outcomes, implicating PWWP2B in cancer biology and genome stability. The function of PWWP2B as a chromatin modifier and DNA repair factor highlights its relevance in transcriptional regulation, thermogenesis, and disease pathophysiology.
