Overview

PX domain-containing protein kinase-like protein (PXK) is a multi-domain protein characterized by a PX domain—which targets it to endosomal membranes—a protein kinase-like domain, and a WASP homology 2 domain[1][8]. PXK regulates membrane protein trafficking, specifically accelerating ligand-induced internalization and degradation of epidermal growth factor receptors via enhanced ubiquitination, and modulates Na,K-ATPase subunits in the brain, impacting electrical excitability and synaptic transmission[1][4][5][6][7]. Although structurally similar to kinases, PXK’s kinase activity is considered atypical, and its main molecular functions relate to protein trafficking and modulation rather than classical phosphorylation signaling[8]. PXK has been associated genetically with systemic lupus erythematosus, but no drugs are currently documented to target PXK directly. Its role in neuronal regulation and immune disease makes it a protein of therapeutic interest and a possible disease biomarker[6].
