PYCARD-AS1 is a long noncoding RNA (lncRNA) transcribed from the opposite strand of the pro-apoptotic gene PYCARD, with dual nuclear and cytoplasmic localization. Mechanistically, PYCARD-AS1 is a key negative regulator of PYCARD, controlling its expression at both epigenetic and translational levels. In the nucleus, PYCARD-AS1 recruits the DNMT1/G9a methylation complex to the PYCARD promoter, facilitating DNA methylation and histone modification, leading to transcriptional silencing. In the cytoplasm, it interacts with PYCARD mRNA to inhibit ribosome assembly, suppressing PYCARD translation. Through coordinated regulation of PYCARD, PYCARD-AS1 influences apoptosis, with implications in cancer biology, notably breast cancer. There are no known drugs targeting PYCARD-AS1 or safety concerns specifically associated with its modulation[1][2][3].
PYCARD-AS1 recruits DNA methyltransferase 1 (DNMT1) and histone methyltransferase G9a to the PYCARD promoter, promoting gene silencing through DNA and histone modification. It also engages in RNA-RNA interaction with PYCARD mRNA, interfering with ribosome assembly and suppressing translation.
