Overview

ASC (Apoptosis-associated Speck-like Protein Containing a CARD), encoded by the PYCARD gene, is a key adaptor protein involved in inflammasome assembly and the regulation of pyroptosis and apoptosis. It contains an N-terminal Pyrin Domain (PYD) and a C-terminal Caspase Recruitment Domain (CARD), which facilitate its interaction with upstream sensor proteins and downstream effectors like procaspase-1. ASC oligomerizes into specks or filaments, serving as a platform for caspase activation and cell death induction. Its expression and function are dysregulated in various diseases, including cancer, infections, and autoimmune disorders.
