Pyridoxal kinase (EC 2.7.1.35) is an essential enzyme that catalyzes the phosphorylation of vitamin B6 vitamers—pyridoxal (PL), pyridoxine (PN), and pyridoxamine (PM)—using ATP as a phosphate donor. This reaction produces their respective 5'-phosphate forms, most notably pyridoxal 5'-phosphate (PLP), which is the biologically active coenzyme form of vitamin B6. PLP acts as a cofactor for over 140 different enzymatic reactions in humans. Dysregulation or mutation of PDXK has been linked to neurological disorders, and overexpression has been associated with cancer cell proliferation.
