Overview

Pyrin and HIN domain-containing protein 1 (PYHIN1, also known as IFIX or Interferon-inducible protein X) is a nuclear protein encoded by the PYHIN1 gene, a member of the HIN-200 family of interferon-inducible proteins[1][3][5][6]. The protein contains an N-terminal pyrin domain (which mediates protein-protein interactions and oligomerization) and one or more C-terminal HIN domains (which can bind DNA)[1][2][6]. PYHIN1 acts as a multifunctional nuclear protein involved in the regulation of the immune response through sensing cytoplasmic and nuclear DNA and promoting the expression of cytokines such as IL-6 and TNFα in response to pro-inflammatory signals[1]. Its tumor suppressor roles are mediated by promoting the degradation of oncogenic regulators such as MDM2 and HDAC1, leading to stabilization of p53 and upregulation of metastasis suppressor genes such as maspin, as well as inducing cell cycle arrest and apoptosis[3][5]. PYHIN1 has been implicated in genetic susceptibility to asthma and other inflammatory diseases, and in restriction of viral replication by binding to viral DNA and suppressing viral gene expression[1][3]. There are no known approved drugs that interact directly with PYHIN1, and it is not currently established as a direct druggable therapeutic target[3].
