Overview

Pyrin domain-containing protein 1 (PYDC1) is a cytosolic adaptor protein that contains only a pyrin domain, distinguishing it from multi-domain inflammasome components[1][3]. It primarily acts as a negative regulator of innate immune signaling by interacting with inflammasome adaptor ASC (PYCARD) and other pyrin domain-containing proteins. Through these interactions, PYDC1 inhibits the activation of NF-κB and pro-caspase-1, resulting in decreased production of pro-inflammatory cytokines such as interleukin-1 beta and suppression of macrophage and granulocyte inflammatory responses[1][3][4]. It is generally considered a cellular checkpoint that limits excessive inflammasome activity and inflammation. No current drugs specifically target PYDC1, and it is not considered a direct therapeutic target, but its role in immune regulation could make it a subject of future research in inflammatory and autoimmune disorders[1][3].
