Pyrin domain-containing protein 5 (PYDC5), also known as pyrin domain-only protein 3 (POP3), is a small regulatory protein expressed in humans and encoded by the PYDC5 gene. It consists of a PYRIN domain and functions as a negative regulator of the AIM2 inflammasome, a multiprotein complex involved in the innate immune response to cytosolic double-stranded DNA, including DNA viruses. PYDC5 inhibits the assembly and activation of the AIM2 inflammasome by binding to AIM2, thus modulating downstream inflammatory responses such as interferon-beta signaling and innate immunity to infections[2][3][7]. PYDC5 belongs to the death domain superfamily (specifically the PYRIN domain subfamily), which includes other regulators of cell death and inflammation, but unlike most members, PYDC5 contains only the PYRIN domain and lacks other effector domains. Currently, PYDC5 is not considered a direct therapeutic target, and no drugs have been identified as acting on this protein.
