It is closely related by sequence homology to pyroglutamyl-peptidase I (PGPEP1), a cysteine-type exopeptidase that removes N-terminal pyroglutamate (pGlu) from peptide substrates[3][11]. PGPEP1L itself is **predicted to possess pyroglutamyl-peptidase (cysteine-type protease) activity**, but its distinct substrate specificity, tissue distribution, and physiological roles are not experimentally well characterized[3][6][11]. The protein is predicted to be **localized in the cytosol** and is involved in general **proteolysis**, potentially participating in the maturation or degradation of bioactive peptides[3][6][11]. No drugs, direct disease associations (other than a potential involvement in chromosome 15q26-qter deletion syndrome), or established clinical uses exist for PGPEP1L at present[3]. Its closest paralog is **PGPEP1**, which has more clearly established roles in neuropeptide processing (e.g., thyrotropin-releasing hormone cleavage) and is a member of the peptidase C15 family[3][4].
