Overview

Pyrroline-5-carboxylate reductase 3 (PYCR3) is a cytoplasmic enzyme that catalyzes the final step of proline biosynthesis, converting pyrroline-5-carboxylate (P5C) to L-proline using NAD(P)H as a cofactor[2][7]. PYCR3 belongs to the pyrroline-5-carboxylate reductase family of oxidoreductases and is primarily involved in the biosynthesis of proline from ornithine[1][2]. The enzyme plays an important role in amino acid metabolism and may influence cell growth and stress responses. Mutations or dysregulation in PYCR3 are associated with rare metabolic disorders such as leukodystrophy (hypomyelinating, type 10) and cutis laxa (autosomal recessive, type IIB), and the gene is expressed in various human cancers[2][5].
