Pyruvate carboxylase (PC) is a biotin-dependent enzyme (EC 6.4.1.1) that catalyzes the ATP-dependent carboxylation of pyruvate to form oxaloacetate, a key intermediate in several metabolic pathways. This reaction is crucial for replenishing oxaloacetate withdrawn from the tricarboxylic acid (TCA) cycle and supports various biosynthetic processes. It plays a critical role in gluconeogenesis, lipogenesis, insulin secretion, and neurotransmitter biosynthesis. PC is regulated by acetyl-CoA (activation) and aspartate/c-di-AMP (inhibition). Mutations in the PC gene are linked with metabolic disorders. Targeting PC is being explored for treating nonalcoholic fatty liver disease and type 2 diabetes.
