Thiamine diphosphate-dependent enzymes (ThDP-dependent enzymes) are a diverse superfamily of proteins that require thiamine diphosphate as a cofactor. These enzymes catalyze various reactions, most notably the formation or cleavage of carbon-carbon bonds, and play essential roles in central metabolism. They are targets for herbicides, antibiotics, anticancer agents, and neuroprotective drugs. Examples include pyruvate decarboxylase, transketolase and DXPS.
