Overview

Pyruvate dehydrogenase E1 subunit alpha 2 is a nuclear-encoded, mitochondrial enzyme subunit, encoded by the autosomal, intronless PDHA2 gene. This subunit is part of the E1 enzyme (α2β2 tetramer) in the pyruvate dehydrogenase complex, which catalyzes the oxidative decarboxylation of pyruvate to acetyl-CoA—a crucial step linking glycolysis and the citric acid cycle. PDHA2 is expressed predominantly in testis tissue due to gene methylation silencing in somatic cells, and it plays a testis-specific role in cellular energy production[2][4]. Regulation occurs at the level of phosphorylation by pyruvate dehydrogenase kinases and is essential for maintaining proper metabolic flux. Dysfunction of the pyruvate dehydrogenase complex is associated with metabolic diseases, and modulation of its activity is considered for treatments of cancer and metabolic disorders, though PDHA2 is less commonly targeted directly compared to PDHA1[1][4].
