Pyruvate dehydrogenase kinase isoform 1 (PDK1) is a mitochondrial enzyme that regulates the activity of the pyruvate dehydrogenase complex (PDC) through phosphorylation. By inhibiting PDC, PDK1 reduces conversion of pyruvate to acetyl-CoA, decreasing entry into the citric acid cycle and promoting conversion to lactate instead. Overactivity of PDK1 has been implicated in cancer cell survival under hypoxic conditions, making it a potential antitumor target. It is also targeted for potential therapies in diabetes and heart disease due to its central role in metabolic flexibility.
Inhibition of PDK1 leads to activation of pyruvate dehydrogenase complex (PDC), promoting the conversion of pyruvate to acetyl-CoA and entry into the citric acid cycle. This shifts metabolism away from glycolysis and lactate production.
