Pyruvate kinase R (PKR) is an isoform of the enzyme pyruvate kinase, which catalyzes the final, rate-limiting step of glycolysis: the conversion of phosphoenolpyruvate (PEP) and ADP to pyruvate and ATP. This reaction is crucial for cellular energy production, especially in tissues that rely heavily on glycolysis, most notably in red blood cells. PKR deficiency leads to impaired ATP generation in red blood cells, resulting in hemolytic anemia. It is predominantly expressed in erythrocytes.
