Overview

Pyruvate-ferredoxin oxidoreductase (PFOR) is a key enzyme in anaerobic metabolism, catalyzing the oxidative decarboxylation of pyruvate to acetyl-CoA and carbon dioxide, while reducing ferredoxin. This reaction is central to energy metabolism in many anaerobic bacteria and archaea, linking glycolysis to various metabolic pathways such as the Wood–Ljungdahl pathway for CO₂ fixation. It requires thiamine pyrophosphate (TPP), three [4Fe–4S] iron-sulfur clusters, and coenzyme A (CoA) as cofactors.
