Overview

Quiescin sulfhydryl oxidase 2 (QSOX2) is an enzyme belonging to the sulfhydryl oxidase family, encoded by the QSOX2 gene, and acts to catalyze the conversion of sulfhydryl groups into disulfide bonds in proteins, a critical step in oxidative protein folding. QSOX2 is secreted into the extracellular space and is periodically expressed throughout the cell cycle, peaking in the G1/S phase and regulated by the transcription factor E2F1. It promotes cell proliferation and survival and is essential for expression of cell division-associated genes and Wnt pathway activators (such as PRRX2 and β-catenin). In cancer, particularly non-small cell lung cancer and neuroblastoma, QSOX2 is overexpressed and associated with poor prognosis and disease progression; its knockdown inhibits proliferation and induces apoptosis in tumor cells. QSOX2 is highlighted as a potential biomarker for assessing tumor burden and response to treatment, but there are currently no specific drugs or therapeutic mechanisms targeting QSOX2 described in the available literature.
