Quinone oxidoreductase NQO1 is a cytoplasmic flavoprotein enzyme that plays a crucial role in cellular defense against oxidative stress and xenobiotic toxicity. It catalyzes the two-electron reduction of quinones to hydroquinones, preventing the formation of semiquinone radicals and reactive oxygen species (ROS). NQO1 is overexpressed in many cancers, and its inhibition can sensitize cancer cells to chemotherapy. Genetic polymorphisms in NQO1 can affect individual susceptibility to toxins and carcinogens.
