Overview

R-spondin 3 (RSPO3) is a secreted protein and member of the R-spondin family that acts as a positive regulator of Wnt signaling pathways. It plays a critical role in embryonic development, stem cell maintenance, angiogenesis, and tumorigenesis. RSPO3 enhances Wnt/β-catenin signaling by binding to LGR4/5/6 receptors on stem/progenitor cells and modulating co-receptors such as LRP6. It also promotes VEGF expression and regulates vascular stability. Its role in cancer is context-dependent, acting as an oncogene in some cases and a tumor suppressor in others.
