Overview

R3H domain containing like (R3HDML) is a protein coding gene that encodes a cysteine-rich, secreted protein predicted to act as a peptidase inhibitor and to bind nucleic acids via its R3H domain[6][1][7]. R3HDML is expressed during skeletal muscle development and regeneration, where it is secreted by satellite cells and is essential for muscle regeneration and myogenic cell differentiation; reduced expression impairs these processes in mouse models[2]. In the kidney, R3HDML is expressed in podocytes, where it acts to inhibit TGF-β-induced p38 MAPK activation, suppressing apoptosis and providing protection during diabetic nephropathy in mice[5]. Its molecular functions include roles as a putative serine protease inhibitor as well as involvement in nucleic acid and polynucleotide binding, likely through the R3H domain, which is conserved and hypothesized to contribute to interactions with single-stranded nucleic acids[1][7][6]. No approved drugs, clinical biomarkers, or safety concerns are currently linked directly to R3HDML.
