Overview

RAAG12 is a primate-restricted N-linked carbohydrate antigen present on multiple membrane-associated proteins. It is highly expressed in colon, gastric, and pancreatic adenocarcinomas, and recognized specifically by the monoclonal antibody RAV12, which is being developed as a therapeutic agent.

Mechanism of action

RAV12 acts as an inhibitor by binding specifically to the N-linked glycotope of RAAG12 expressed on cancer cells' membranes. This interaction can block receptor function (e.g., IGF1R), disrupt signaling pathways essential for tumor growth/survival, induce oncotic cell death directly through membrane perturbation or immune effector mechanisms such as ADCC/CDC.
