Overview

Rab11 family-interacting protein 3 (RAB11FIP3) is a cellular adaptor protein involved in membrane trafficking and vesicle transport within cells. It acts as a downstream effector for Rab11 GTPase, regulating endocytic recycling, membrane delivery during cell division (cytokinesis), and intracellular ciliogenesis. RAB11FIP3 is critical for maintaining the integrity and function of the endosomal recycling compartment (ERC) and facilitates the movement of endosomes along microtubules by linking Rab11 to the dynein motor complex. RAB11FIP3 operates through direct interaction with Rab11A and forms complexes with cytoskeletal motor proteins and the exocyst, orchestrating the inward trafficking of vesicles for membrane recycling, division completion, and ciliary assembly. Its functional disruption leads to defects in cytokinesis, abscission, and cell proliferation, underlying its roles in diseases such as congenital muscular dystrophy, Brugada syndrome, and potential involvement in cancer cell motility. It is not classified as a receptor, enzyme, or transporter but rather functions as an adaptor protein in the Rab GTPase trafficking network.
