Overview

Rab11 family-interacting protein 5 (RAB11FIP5) is a cytosolic adaptor protein involved in regulating protein trafficking from apical recycling endosomes to the plasma membrane[1][2][9]. It binds to small GTPases of the Rab family—particularly RAB11A and RAB25—and helps coordinate slow recycling of membrane proteins, such as endocytosed receptors, by tethering vesicles to cytoskeletal motors like kinesin II[2]. RAB11FIP5 plays important roles in exocytosis, such as insulin granule release, and in specialized processes like maintaining neuronal AMPA receptor localization and cell polarity, as well as facilitating cellular responses to acidic environments via regulation of V-ATPase[2][9]. It is associated with neurodevelopmental and autoimmune pathologies, but is not considered a direct therapeutic target, nor are there approved drugs interacting with it[1][6][8].
