Rab15 effector protein (REP15) is a compartment-specific effector that interacts with active (GTP-bound) Rab GTPases, primarily Rab15, and also Rab3 paralogs and Rab34[2][3]. REP15 localizes to the endocytic recycling compartment (ERC), where it colocalizes with Rab15 and Rab11, regulating the recycling of the transferrin receptor, a process essential for iron homeostasis and membrane trafficking. REP15 does not contain known functional or membrane domains seen in other Rab effectors and adopts a unique globular structure[1][2]. Functionally, manipulation of REP15 expression (either overexpression or knockdown) disrupts transferrin receptor recycling from the ERC without affecting the rapid recycling from sorting endosomes, highlighting its compartment specificity[1]. In glioblastoma cells, REP15 depletion impairs cell proliferation and migration, implying a potential but not yet fully defined role in oncogenesis[2][4]. No drugs are currently known to target REP15 directly, and there are no established mechanisms of drug action, biomarker utility, or specific therapeutic safety concerns associated with this protein. REP15's principal significance is as a basic cell biology regulator rather than a current clinical target.
