Overview

Rabenosyn-5 is a cytosolic effector protein that plays a critical role in **early endosomal membrane trafficking** by acting as a direct binding partner for active (GTP-bound) Rab5, a small GTPase involved in endocytosis[3][1][2]. Structurally, Rabenosyn-5 contains an N-terminal C2H2 zinc finger, a FYVE domain that binds phosphatidylinositol-3-phosphate (PI3P), and distinct regions that interact with multiple Rab proteins, including Rab4 and Rab5[3][2][1]. Rabenosyn-5 also forms a complex with hVPS45, functionally linking Rab5-dependent endosome docking and fusion events with vesicle trafficking machinery, and acts as a central organizer for the recruitment of other trafficking proteins[3]. The protein is essential for efficient endosomal fusion and protein sorting but is not currently recognized as a direct therapeutic target, and there are no known approved drugs targeting this molecule[3][4]. Overexpression of Rabenosyn-5 can selectively inhibit the processing of lysosomal enzymes (e.g., cathepsin D), suggesting a specific and distinct functional role within endosomal sorting pathways compared to other Rab effectors such as EEA1[3].
