Overview

Rabies lyssavirus glycoprotein G is the sole surface-exposed protein on the rabies virus envelope, forming trimeric spikes that recognize and bind host cell receptors, triggering endocytosis and subsequent low pH-dependent membrane fusion to facilitate viral entry. It is the crucial antigen for the generation of virus-neutralizing antibodies and is the primary target for rabies vaccines and therapeutic antibodies. The G protein undergoes reversible conformational changes between prefusion and postfusion states, influencing antigenic exposure and immune recognition. Variants of G protein can affect pathogenicity, immunogenicity, and vaccine responses. Neutralization of the G protein is the key mechanism for clinical protection from rabies infection.
