Rac family small GTPase 1 (RAC1), historically also known as TC25, is a critical signaling protein belonging to the Rho family of small GTPases. It functions as a molecular switch, cycling between an inactive GDP-bound state and an active GTP-bound state to regulate a wide array of cellular processes, most notably actin cytoskeleton remodeling, which drives cell motility, adhesion, and shape changes. Beyond its structural roles, RAC1 is a key mediator of signal transduction pathways that control cell cycle progression, gene expression, and the production of reactive oxygen species via the NADPH oxidase complex. In the context of human disease, RAC1 is frequently dysregulated or mutated in various cancers, where it promotes tumor invasion, metastasis, and resistance to therapy. For instance, the RAC1 P29S "fast-cycler" mutation is a known driver in cutaneous melanoma. Because of its central role in oncogenic signaling and cell migration, RAC1 has become a significant therapeutic target. Current drug development efforts focus on small-molecule inhibitors that prevent its activation by Guanine Nucleotide Exchange Factors (GEFs) or block its interaction with downstream effectors like PAK1, aiming to halt cancer progression and address other conditions such as inflammatory and neurodegenerative disorders.
