Overview

Rac GTPase-activating protein 1 (RACGAP1) is an enzyme encoded by the RACGAP1 gene in humans and acts as a GTPase-activating protein (GAP) for Rho family GTPases, stimulating their GTP hydrolysis and thus negatively regulating their activity. RACGAP1 is a central component of the centralspindlin complex, which is critical for cytokinesis, the process by which dividing cells physically separate. It interacts with the protein KIF23 and is required for cleavage furrow ingression and abscission. RACGAP1 plays key roles not only in cytokinesis but also in cell growth, differentiation (including erythropoiesis), and signal transduction. Mutations in RACGAP1 are associated with congenital dyserythropoietic anemia type IIIB, and its overexpression is implicated in various cancers such as breast, gastric, and colorectal cancer. The protein also participates in mitochondrial quality control in cancer cells and is essential for embryonic development[1][2][3].
