Overview

The Rac1–PAK1–p38 MAPK signaling pathway is a key intracellular cascade that transmits signals from extracellular stimuli to downstream cellular responses. Rac1, a member of the Rho family of small GTPases, activates PAK1, a serine/threonine kinase, which in turn can activate the p38 MAPK subfamily. This pathway regulates diverse processes including cell proliferation, apoptosis, inflammation, cell cycle progression, and gene expression. Upstream and downstream effectors integrate environmental and genetic cues, making the pathway central to normal cell biology and numerous diseases, especially cancer and inflammatory conditions. The individual molecules within the pathway (Rac1, PAK1, p38 MAPKs) are considered drug targets, but the pathway itself is not a single canonical target[1][2][3][4].
