RAD51 antisense RNA 1 (RAD51-AS1), also known as TODRA, is a long non-coding RNA transcribed from the opposite direction of the RAD51 gene locus. RAD51-AS1 plays regulatory roles in cancer biology by modulating RAD51-dependent DNA repair, influencing cell cycle progression, and apoptosis. The functional outcomes of RAD51-AS1 are highly context-dependent: in some cancers, such as ovarian and breast cancer, RAD51-AS1 is reported to promote cell proliferation, inhibit apoptosis, and enhance DNA repair, leading to poorer prognosis. In contrast, in hepatocellular and colorectal cancers, higher expression or enforced expression of RAD51-AS1 inhibits RAD51 translation, reduces cell proliferation, and sensitizes cells to chemotherapy and radiotherapy. Its mechanisms are believed to involve formation of double-stranded RNA with complementary sense transcripts, either stabilizing or downregulating the corresponding protein, depending on the cellular context. No direct small molecule or drug modulators of RAD51-AS1 are reported so far[4][5].
