RAD51-associated protein 1 is a structure-specific DNA-binding protein that facilitates homologous recombination by physically interacting with RAD51 and DNA intermediates, enhancing the RAD51-mediated formation of joint molecules (D-loops) during DNA repair. It possesses two distinct DNA-binding domains, both essential for its role in promoting efficient DNA repair and maintaining genome integrity. RAD51AP1 is required for cellular resistance to DNA double-strand breaks, operates both in somatic and meiotic recombination, and has a key role in alternative lengthening of telomeres in cancer cells. Mutations or dysregulation of RAD51AP1 are associated with genomic instability and disease, especially cancer and hereditary anemias[1][2][4].
Agents or molecules that inhibit RAD51AP1 function may impair homologous recombination repair, sensitize cancer cells to DNA-damaging agents, and promote genome instability. Synergistic action with other DNA repair homology-directed pathway components[2][4].
