RAD51B antisense RNA 1 (RAD51B-AS1) is a 537 nucleotide long non-coding RNA located predominantly in the nucleus of ovarian cancer cells. It is upregulated in highly metastatic ovarian cancer cell lines and tissues, where it promotes malignant biological behaviors including cell proliferation, metastasis, and resistance to anoikis (a form of apoptosis associated with cell detachment). RAD51B-AS1 exerts its effects primarily by positively regulating the expression of its sense gene RAD51B, thereby activating the Akt/Bcl-2 signaling pathway. Its expression correlates with poor prognosis in ovarian cancer, making it both a potential molecular biomarker and a therapeutic target. The mechanisms by which RAD51B-AS1 regulates gene expression are not fully elucidated, but may include interactions with proteins, DNA, or other RNAs at the pre- and post-transcriptional levels. No drugs are currently approved to target RAD51B-AS1, but silencing approaches have shown promise in experimental models.
For experimental antisense oligonucleotides: Gene silencing through direct targeting of RAD51B-AS1, resulting in decreased cell proliferation and increased apoptosis in ovarian cancer cell lines. For hypothetical therapies: Modulation of lncRNA activity affecting the Akt/Bcl-2 pathway and downstream oncogenic processes.
