Ral guanine nucleotide dissociation stimulator like 4 (RGL4) is a protein-coding oncogene that encodes a guanine nucleotide exchange factor (GEF) similar to Ral guanine nucleotide dissociation stimulator. It activates small GTPases—specifically RalA and RalB—by promoting the exchange of GDP for GTP, playing a role in signal transduction from Ras to downstream pathways such as the Ras-Raf-MEK-ERK cascade. Increased expression results in protein translocation to the cell membrane and enhanced activation of signaling pathways involved in cell proliferation and oncogenesis. RGL4 is related to the Ras effector family and is implicated as a molecular contributor to cancer biology, but there are currently no direct interacting drugs or well-established biomarker or safety concern associations.
