RalA-binding protein 1 (RALBP1) is a multifunctional effector protein acting downstream of the small GTPases RALA and RALB[1][2][6]. It integrates Ras and Rho family signalling by serving as both a GTPase-activating protein (GAP) for Rho family members (such as CDC42 and RAC1) and a scaffold protein facilitating mitosis-specific kinase signaling[1][2]. RALBP1 plays essential roles in receptor-mediated endocytosis, cell migration, mitochondrial fission, and in mediating the cellular response to metabolites and toxins by active ATP-dependent transport of glutathione conjugates and xenobiotics, including chemotherapeutic agents like doxorubicin[2][6]. Overexpression or dysregulation of RALBP1 is linked to cancer cell survival, multidrug resistance, and potentially tumor progression[2][3].
