RAMP2 antisense RNA 1 (RAMP2-AS1) is a long non-coding RNA (lncRNA) located on the antisense strand of the RAMP2 gene. It acts as a regulatory RNA that influences a wide range of cellular processes, particularly in endothelial cells, where it modulates angiogenesis, endothelial homeostasis, and cellular aging. RAMP2-AS1 positively regulates RAMP2 mRNA by stabilizing it through an interaction with the RNA-binding protein TIA1, and may also act in cis to control RAMP2 transcription. Broader transcriptomic analyses indicate that RAMP2-AS1 inhibition impacts key pathways in endothelial cells, including those involved in angiogenesis, apoptosis, immune and inflammatory signaling, extracellular matrix remodeling, and amino acid transport. In several cancers, including non-small cell lung cancer and glioblastoma, RAMP2-AS1 exhibits tumor suppressor activity, with overexpression reducing proliferation, invasion, and promoting apoptosis. Downregulation of RAMP2-AS1 is associated with poorer prognosis in these cancers, and it may serve as a biomarker for disease outcome. There are no known drugs that directly target RAMP2-AS1, and its safety as a therapeutic target has not been established[1][2][4][5][6][7][8].
