Overview

Ran-binding protein 3 is a predominantly nuclear transport accessory protein encoded by the RANBP3 gene. It binds Ran-GTP and functions as a key cofactor for CRM1/XPO1-mediated nuclear export, stabilizing export complexes and regulating the nucleocytoplasmic transport of proteins such as SMAD2, SMAD3, and β-catenin, thus acting as a negative regulator of TGF-beta signaling. RANBP3 also enhances the guanine nucleotide exchange activity of RCC1, and its nuclear import is mediated preferentially by importin-α3. The protein is broadly expressed and its dysfunction is implicated in processes relevant to cancer progression and certain genetic disorders, but is not currently considered a direct therapeutic target[1][2][3][4][5].
