Ran-binding protein 3-like (RANBP3L) is a protein containing a conserved Ran-binding domain and functions as a nuclear export factor specific for dephosphorylated SMAD1/5/8 in the BMP signaling pathway[1][3]. RANBP3L plays a critical role in terminating BMP signaling and regulating the differentiation of mesenchymal stem cells by mediating the nuclear export of SMADs in a Ran-dependent manner. In the kidney, RANBP3L is regulated by the transcription factor NFAT5 in response to hyperosmotic stress, supporting renal epithelial integrity and acting as a putative tumor suppressor, as its loss promotes a tumorigenic phenotype associated with renal cell carcinoma, especially the clear cell subtype[2][3]. Notes: - RANBP3L is not a typical therapeutic target (such as a receptor, enzyme, ion channel, or transporter), but a nuclear transport cofactor/adaptor. - No clinically approved drugs are known to directly interact with RANBP3L, nor are there known small-molecule modulators. - Its expression level (not function as a drug target) may be of prognostic significance in renal cancer[2]. - The gene is sometimes listed as FLJ25422 in older literature[3].
